2366 After the Borg attack on Earth the Defiant project is started. The USS Defiant is planned as a experimental prototype equippest with the most recent weapons and propulsion technologies in order to be better prepared in case of another Borg incident. 
2366-69 The USS Defiant is constructed in the Utopia Planitia Fleet Yards in the orbit of Mars under the command of Cmd. Benjamin Sisko. 
2370 Stardate 47538.5 The USS Defiant is commissioned in the Antares Fleet Yards. 
2375 Stardate 52584 The USS Defiant is destroyed in the Chin'toka system by a Breen vessel while defending the location against Dominion forces.

2371 Stardate 48212.4 Due to the increasing threat of the Dominion the Defiant is permantently ordered to Deep Space Nine, near the gateway to the Gamma Quadrant, and put unter the command of Cmd. Benjamin Sisko. She is equipped with a Romulan cloaking device, which, according to the treaty with the Romulans, may only be used in the Gamma Quadrant and under the condition that all gathered information are shared. 
On her first mission, the Defiant flies to the Gamma Quadrant to find the home planet of the founders. 
2373 Stardate 50893 Under the command of Lt. Cmd. Worf, the USS Defiant is a part of the defensive fleet of the Federation that tries to stop the Borg cube invading Federation space. She is involved in the attack in the Typhon sector as well as in Sector 001, where they finally suceed in destroying the cube. In battle, the Defiant is heavily damaged, and the entire crew has to be evacuated to the Enterprise-E. However, soon afterwards the ship is salvaged and repaired within few weeks. 
December After the attack of the Dominion on DS9 and the beginning of the war, the Starfleet crew of the station is evacuated with the Defiant. Afterwards, the ship joins the 6th fleet in order to help defending the Alpha Quadrant. 
2374 February The USS Defiant is ordered to Starbase 375, where Captain Sisko is relieved of command and, together with his commanding officers, is transferred to the Jem'hadar ship that was captured one year ago. 
May The Defiant, that is again under the command of Captain Sisko in the meantime, is the vessel that leads the retake of Deep Space Nine. 
December During the invasion of the Cardassian Chin'toka system by mixed tactical forces of the Federation, the Klingons and Romulans the Defiant leads the vessels of the Federation. 
2375 Stardate 52584 During the defense of the Chin'toka system, that was captured by the Federation one year ago and is now attacked by a Dominion/Breen fleet, the USS Defiant is disabled by a energy draining weapon of the Breen. Since propulsion systems, shields, weapons and emergency force fields fail, the Defiant is defenselessly exposed to the attacks, so that the crew has to use the escape pods and abandon the ship. Shortly afterwards, the Defiant is destroyed.
